LONDON (Reuters) ? A class of generic AIDS drugs often used to treat HIV in Africa and other poor regions can cause premature aging and lead to age-related illnesses such as heart disease and dementia, scientists said on Sunday.
In a study in the journal Nature Genetics, British researchers found that the drugs, known as nucleoside analog reverse-transcriptase inhibitors, or NRTIs, damage DNA in the patient's mitochondria -- the "batteries" that power cells.
The scientists said it was unlikely that newer cocktails of AIDS drugs made by firms like Gilead, Merck, Pfizer and GlaxoSmithKline would inflict similar levels of damage, since they are thought to be less toxic to mitochondria. But more research is needed to be certain.
"It takes time for these side effects to become apparent, so there is a question mark about the future and whether or not the newer drugs will cause this problem," Patrick Chinnery of the Institute of Genetic Medicine at Newcastle University said in a telephone interview. "They are probably less likely to, but we don't know because we haven't had time to see."
The findings do however help explain why HIV-infected people treated with older antiretroviral AIDS drugs sometimes show advanced signs of frailty and diseases such as heart disease and dementia at an early age, the researchers said.
"The DNA in our mitochondria gets copied throughout our lifetimes and, as we age, naturally accumulates errors," said Chinnery, who led the study.
"We believe these HIV drugs accelerate the rate at which these errors build up. So over the space of, say, 10 years, a person's mitochondrial DNA may have accumulated the same amount of errors as a person who has naturally aged 20 or 30 years."
NRTI drugs -- the best known of which is AZT, also known as zidovudine and originally developed by GSK -- were a big advance in HIV treatment when they first emerged in the late 1980s. They extended patients' lives and helped make HIV a manageable chronic disease rather than the death sentence it once was.
Concerns about toxicity of NRTIs, particularly with long-term use, mean the drugs are now less commonly used in wealthy countries where they have been replaced by newer more expensive combination AIDS drugs with fewer side-effects.
But in poorer countries, where access to cheaper generic medicines is often the only option for HIV patients to get treatment, NRTIs are still relatively widely used.
An estimated 33.3 million people worldwide had the human immunodeficiency virus (HIV) that causes AIDS in 2009, according to the latest United Nations data, and 22.5 million of those live in Africa.
"These drugs may not be perfect, but we must remember that when they were introduced they gave people an extra 10 or 20 years when they would otherwise have died," said Brendan Payne of Newcastle's Royal Victoria Infirmary, who also worked on the study.
"In Africa, where the HIV epidemic has hit hardest and where more expensive medications are not an option, they are an absolute necessity."
For their study, Chinnery's team studied muscle cells from HIV-infected adults, some of whom had previously been given NRTIs. They found that patients who had been treated with NRTIs -- even as long as 10 years previously -- had damaged mitochondria similar to that of a healthy older person.
The researchers are now looking at ways to repair or stall some of the damage caused by the drugs and say they believe that focusing on exercise -- which appears to have a beneficial effect on patients with mitochondrial diseases -- may help.

Chancellor Merkel insists that Greece must swallow more austerity to get EU help EU leaders have urged all Greek politicians to support new spending cuts and tax hikes, saying there is no alternative if debt-laden Athens is to qualify for a second massive bail-out.
The second rescue is being negotiated in Brussels. It is expected to be about 120bn euros (£107bn; $171bn).
"There will be a new programme for Greece, on which the Greek parliament will have to vote next week," said Germany's Chancellor Angela Merkel.
The UK says it will not contribute.
EU leaders have begun a second day of summit talks in Brussels dominated by the Greek debt crisis, which threatens the stability of the 17-nation eurozone.
"In the case of Britain I sought assurances that Britain wouldn't be pulled into a eurozone package for Greece, and I've received those assurances," UK Prime Minister David Cameron said.
Ms Merkel reiterated: "We will do everything to stabilise the euro."
She spoke of the Greek opposition needing to "fulfil its historic responsibilities".
The BBC's Matthew Price in Brussels says Europe's exasperation with Greece's economic woes is all too clear.
On Thursday night the EU leaders called on "all political parties in Greece to support the programme's main objectives", saying national unity was a "prerequisite for success".
Our correspondent says it was a direct challenge to the Greek opposition, which says it will not support the budget cuts when they are put to parliament next week.
For weeks Athens has been gripped by huge street protests and strikes by Greeks opposed to the tough bail-out conditions. Many of them think Greece is being sacrificed to save the euro.
Greece's government is proposing additional spending cuts worth 28bn euros over five years.
Herman Van Rompuy: "Required additional funding will be financed through both official and private sources"
If these are passed then Greece will get its next 12bn-euro instalment from the current 110bn euro bail-out package from the EU and International Monetary Fund.
Without this money Greece will default on its next loan payments due in mid-July.
However, many economists think it will be difficult for Greece to avoid default at some time in the future.
European Council President Herman Van Rompuy said the EU was also moving to make it easier for Greece to access EU development funds to help boost its economy.
Earlier, Luxembourg Prime Minister Jean-Claude Juncker urged Athens to meet its commitments under the bail-out programme.
"All conditions must be met," said Mr Juncker, who also chairs the group of 17 eurozone finance ministers. "You can't let anyone believe there is a Plan B. If Greece does what it has to do, we will do what we have to do."
Greek Prime Minister George Papandreou said his government was committed to pushing the austerity plans through parliament next week.
"Greece is committed, strongly committed, to continue a very important programme for major changes, radical changes, to make our economy viable," he said.

It turns out that students and parents who have turned down traditional education still long for a little ?Pomp and Circumstance.? As the number of homeschoolers in the U.S. balloons to 2 million children, high school graduation ceremonies are the new hot trend in non-traditional education.
This June, a group of students donned capes and listened to speeches in what organizers called the ?very first South Florida home-school graduation.?

?I was getting all down when I didn't think he'd have a graduation,? one proud mother of the ?home-school class of 2011? told the New York Times. ?I wanted to see him walk and have the cap and gown and the pictures.?
The Florida group had just 26 students, but some organizations are holding mass convocations. Christian groups like the Home Educators Association of Virginia offer graduation ceremonies for hundreds of homeschoolers at their annual conventions.
There are plenty of accoutrements for the proud parents too. Online retailers catering to the burgeoning home-school market sell everything from class rings to ?Proud of Our 2011 Home-School Graduate? yard signs.
Some students are reluctant to participate, in part because their classmates are strangers. I guess I can see how that would be the case since like I said before, their efforts were already focused on other platforms. I?m sure there are many BlackBerry users who were faithful to the Seesmic app but don?t worry, this isn?t the only Twitter app for BlackBerry and it?s definitely not as fully featured as others which could be one of the reasons they quit. Just saying.

Though they can easily be adapted to fit the business practices of smaller companies, ERP systems were originally designed with large enterprises in mind. It will come as no surprise, then, to discover that the U.S. Military is currently engaged in a lengthy and expensive ERP implementation that is expected to automate all Defense Department systems across all divisions of the Armed Forces.
The larger an organization, the longer it will take to successfully customize and implement an ERP system. According to DefenseSystems.com, the military's implementation is actually taking place across nine ERP platforms originated from the Department of Defense's ARPANET (Advanced Research Projects Agency Network) decades ago. The depth and breadth of the U.S. Military's ERP installation offers many lessons in ERP planning and implementation that can be taken to heart by companies of any size.
The military's ERP system is being segmented into three major categories: Air Force, Navy and Army. This makes sense, given the different natures of these organizations. An entirely-unified system is probably a bit much, considering that the entire budget for the Department of Defense is $663 billion. However, further divisions might make for too much fragmentation. When one considers that the Marines are ground troops under the purview of the Navy, things begin to get complicated. Too much complexity (especially on the vast scale that these systems will exist on) can draw out an implementation period to the point where a system is obsolete before it goes online.
Large ERP implementations (especially those that are paid for by enormous amounts of taxpayer funds) require a great deal of planning. Consequently, it is probably not a good idea to try and implement everything at once. However, it is also not desirable to start and finish one system at a time because it will be more difficult to link them all up. A balance must be struck between synergistic automation and rapid deployment. Therefore, a hard look must be taken at the divisions within a proposed ERP system. For example, the U.S. Military's Naval systems are much further along than its Air Force platforms. The reasoning behind this is that the Navy is operating with legacy systems that are much more outdated and therefore require a first crack at the updated software. The Air Force can stand to wait, though work on those systems began in earnest before Naval programs were complete.

WASHINGTON (Reuters) ? A monster black hole shredded a Sun-like star, producing a strangely long-lasting flash of gamma rays that probably won't be seen again in a million years, astronomers reported on Thursday.
That is definitely not the norm for gamma ray bursts, energetic blasts that typically flare up and end in a matter of seconds or milliseconds, often the sign of the death throes of a collapsing star.
"This is truly different from any explosive event we have seen before," said Joshua Bloom of the University of California-Berkeley, a co-author of research on the blast published in the journal Science.
Initially spied on March 28 by NASA's Swift spacecraft, which is trolling the universe for gamma ray bursts, this particular flash has lasted more than two months and is still going on, Bloom said in a telephone interview.
What makes this even stranger is that the black hole, located in the constellation Draco (The Dragon) about 4 billion light years, or 24 sextillion miles (38.62 sextillion km) -- 24 followed by 21 zeroes -- from Earth, was sitting quietly, not eating much, when a star about the mass of our Sun moved into range.
"We have this otherwise dormant black hole, not gobbling up an appreciable amount of mass, and along comes this star which just happens to be on some orbit which puts it close to the black hole," Bloom said.
FEEDING FRENZY
"This was a black hole which was otherwise quiescent and it sort of has an impulsive feeding frenzy on this one star," he said.
Bloom figures this may happen once per black hole per million years.
This kind of behavior is different from what active black holes generally do, which is to suck in everything their vast gravity can pull in, even light. Most galaxies, including our Milky Way, are thought to harbor black holes in their hearts.
Black holes are invisible, but astronomers can infer their existence because the material they pull in lights up before it gets sucked in.
In this case, though, the black hole feasted on one star -- about the same mass as our Sun -- with such relish that it tore the star apart before gulping it down. As it did so, the black hole emitted powerful gamma ray jets from its center as bits of the dying star were turned into energy.
The black hole's gravitational pull was so great that it exerted what's called a tidal disruption on the passing star.
Astronomers could use this observation to help them learn more about how black holes grow, Bloom said.
"We still don't understand how black holes and the universe grow," he said. "We think most black holes start off as being no more than the mass of our Sun ... How they go from 10 solar masses to a billion solar masses is critical."
There is a strong connection between the mass of black holes and the mass of the galaxies that host them, with black holes feeding on gas and stars that come near.
